چکیده
Abstract The a priori T cell repertoire and immune response against SARS-CoV-2 viral antigens may explain the varying clinical course prognosis of patients having mild COVID-19 infection as opposed to those developing more fulminant multisystem organ failure associated mortality. Using novel SARS-Cov-2-specific artificial antigen presenting (aAPC), coupled with rapid expansion protocol (REP) practiced in tumor infiltrating lymphocytes (TIL) therapy, we generate an catalytic quantity Virus Induced Lymphocytes (VIL). receptor (TCR)-specific aAPCs carrying co-stimulatory molecules major histocompatibility complex (MHC) class-I immunodominant peptide-pentamer complexes, expand virus-specific VIL derived from peripheral blood mononuclear cells (PBMC) convalescent up 1000-fold. This is achieved clinically relevant 7-day vein-to-vein time-course potential adoptive therapy (ACT) for COVID-19. We also evaluate this approach other pathogens using Cytomegalovirus (CMV)-specific donors control. Rapidly expanded are enriched virus antigen-specificity show activated, polyfunctional cytokine profile effector memory phenotype which contribute robust response. Virus-specific can be delivered allogeneically via MHC-typing patient human leukocyte (HLA)-matching provide pragmatic treatment large-scale therapeutic setting. These data suggest that represent option warrants further investigation armamentarium possible future pandemics.

COVID-19: A New Virus as a Potential Rapidly Spreading in the Worldwide
Covid-19 is a novel virus with high affinity to spread in the community. In December 2019, it was first identified in Wuhan, China. The symptoms are non-specific, so fever, cough, dyspnea, are prominent features. Respiratory failure and mortality have also been reported. The most common lung CT scan findings are bilateral ground glass opacities.

In 2019 a newly emerged coronavirus was detected by the Center for disease control (CDC) in China. Nucleic acid sequencing from nose and throat swab samples of patients revealed that it was like severe acute respiratory syndrome coronavirus (SARS-CoV). World Health Organization (WHO) named it coronavirus disease 2019 (COVID-19) and reported more than 100000 positive tests until March 2020 for C...
متن کاملInfluenza virus-infected epithelial cells present viral antigens to antigen-specific CD8+ cytotoxic T lymphocytes.
We have investigated the mechanisms involved in the clearance of viral infection at the epithelium level by analyzing the activity of influenza virus-specific cytotoxic T lymphocytes (CTL) against virus-infected CMT-93 intestinal epithelial cells. Epithelial cells infected with live influenza virus effectively present viral antigens and were lysed by both homotypic and heterotypic influenza vir...
متن کاملAmmonium Chloride as a Potential Candidate for the Treatment and Controlling of Covid-19
Coronaviruses, pathogens with a zoonotic potential, are positive sense single-stranded RNA viruses. SARS Coronavirus-2, the cause of Covid-19 infection, is from the betacoronavirinea subfamily, which has close genomic and proteomic similarity to SARS Coronavirus-1(1). Given the genomic proximity of these two viruses, studies on SARS Coronavirus-1 can be used to control or detect SARS Coronaviru...
